Dietary supplementation with a concentrate enriched in milk fat globule membranes (MFGMs) and extracellular vesicles (EVs) promotes the accretion of distinct sphingolipid profiles in aged sedentary rats, including reductions in plasma ceramides associated with elevated cardiovascular risk. Over a 25-week intervention, the MFGM/EV-rich supplement significantly improved lipoprotein particle profiles marked by reduced triglyceride-rich lipoproteins and exhibited a general lipid-lowering effect across plasma and hepatic tissues. These changes were accompanied by evidence of enhanced lipid turnover and metabolic flux, indicating systemic improvements in lipid metabolism. Collectively, these findings highlight the potential of MFGM/EV-based nutraceuticals as a non-pharmacological strategy to mitigate age-related metabolic dysfunction.
