PKP2: A New Therapeutic Target in Alphavirus Management

The study evaluates how the bacterial effector protein IpaH2, an E3 ubiquitin ligase produced by Shigella flexneri, influences viral infection mechanisms within host cells. The researchers identified plakophilin-2 (PKP2), a cellular adhesion and signaling protein, as a previously unrecognized host restriction factor that limits the replication of Sindbis virus. Through its ubiquitin ligase activity, IpaH2 targets PKP2 for degradation, thereby weakening the cell’s antiviral defense and enhancing viral replication. These findings reveal a novel interaction between bacterial virulence factors and host antiviral pathways, highlighting PKP2 as an important component of innate cellular defense and providing new insights into how microbial pathogens can manipulate host ubiquitin signaling networks to influence viral susceptibility.
