Osteogenic Potential of Limosilactobacillus fermentum GBE18 Cell-Free Supernatant

The study evaluated the cell-free supernatant (CFS) of Limosilactobacillus fermentum GBE18 for its potential to promote bone formation using MC3T3-E1 preosteoblast cells. The probiotic-derived CFS contains soluble metabolites and bioactive compounds released by L. fermentum GBE18, which enhanced osteogenic activity in the cells. Treatment increased markers associated with osteoblast differentiation and mineralization, with the effects linked to activation of the Wnt/β-catenin and PI3K/Akt signaling pathways, two important pathways involved in osteoblast proliferation, differentiation, and bone matrix formation. The CFS promoted β-catenin signaling and Akt phosphorylation, supporting downstream osteogenic responses and increasing the expression of bone-related markers. CFS from Limosilactobacillus fermentum GBE18 has promising osteogenic potential and may represent a postbiotic-based approach for supporting bone formation and skeletal health.
