In an observational study of 3,602 infertile women with PCOS, treatment with myoinositol (2 × 2000 mg/day) plus folic acid (2 × 200 μg/day) for a mean 10.2 weeks restored ovulation in 70% of participants and produced 545 pregnancies (15.1% overall). Concomitant drugs were rare, one twin pregnancy occurred, and no relevant adverse effects were reported.
A hormonal subanalysis (n=32) showed significant improvements after 12 weeks: total testosterone fell from 96.6 ng/ml to 43.3 ng/ml and progesterone rose from 2.1 ng/ml to 12.3 ng/ml (p < 0.05). Results suggest a safe, effective alternative for PCOS-related infertility with outcomes comparable to metformin.
